Jumpstart your professional career in Implementation Science research working with behavioral intervention researchers at the University of Massachusetts Medical School.  This 2-year post-doctoral fellowship focuses on implementation science research in HIV.  The post-doctoral fellow will work on a NIH-funded national implementation study in The Bahamas and an mHealth PrEP uptake project in Thailand. The goal of the national implementation study is to develop theory-driven implementation strategies to increase sustained teacher implementation fidelity of youth evidence-based intervention, and thereby increase the public health impact of evidence-based interventions in schools. The mHealth project aims to adapt effective mHealth interventions to improve uptake and adherence of the HIV pre-exposure prophylaxis (PrEP) in Thai young MSM. Development of novel ZFP-based gene therapy approaches for monogenic disorders.   The Wolfe laboratory will be filling a postdoctoral position to develop new gene therapy reagents for the treatment of monogenic disorders in collaboration with other laboratories within UMMS. These efforts will focus on the creation of novel Cys2His2 zinc finger protein-based therapeutics for delivery to affected cell types.  This project will focus on:   1) The development of Cys2His2 zinc finger proteins (ZFPs) with novel DNA binding specificity to target specific (therapeutic) genomic loci.   2) The application of these ZFPs in cell culture and in vivo models of disease to abrogate disease-associated phenotypes.     These studies will expose the successful candidate to cutting-edge ZFP design and selection approaches, as well as in vitro and in vivo delivery approachs.  The successful candidate will also learn cutting edge deep-sequencing approaches to monitor the genome-wide binding locations and impact of the ZFPs on the genome and transcriptome. The successful candidate will have the opportunity to participate in meetings/conferences associated with gene therapy, which will broaden their exposure to cutting edge research within the field. In addition, the successful candidate will learn to utilize CRISPR-Cas9 and CRISPR-Cas12a nuclease technologies that are being developed in the Wolfe laboratory. Candidates should have a Ph.D. in biology, biochemistry, chemistry or a related biomedical science. In addition, the potential candidate must have strong oral and written communication skills and be eager to present research results at national meetings. Development of ex vivo genome editing approaches for hematopoietic disorders.   The Wolfe laboratory will be filling a postdoctoral position to develop improved genome editing reagents and approaches for sickle cell disease, beta-thalassemia and other hematopoietic disorders with the goal of translating these reagents to clinical trials.  This project will focus on:   1) The development of CRISPR editing reagents with high activity and specificity for ex vivo genome editing in human CD34+ HSPCs.   2) The utilization of unbiased genome-wide techniques for assessing off-target events and other genomic alterations potentially associated with genome editing.   3) The translation of the developed genome editing reagents to clinical application.   These studies will expose the successful candidate to cutting-edge CRISPR/Cas nuclease, base editor and prime editor engineering approaches, and the ex vivo delivery of these reagents to CD34+ HSPCs.  The successful candidate will also learn cutting edge deep-sequencing approaches to evaluate nuclease-based collateral damage to the genome. The successful candidate will have the opportunity to participate in meetings/conferences associated with gene therapy, which will broaden their exposure to cutting edge research within the field. In addition, the successful candidate will learn about Cys2His2 zinc finger protein gene regulation technologies that are being developed in the Wolfe laboratory.
