Development of ex vivo genome editing approaches for hematopoietic disorders.   The Wolfe laboratory will be filling a postdoctoral position to develop improved genome editing reagents and approaches for sickle cell disease, beta-thalassemia and other hematopoietic disorders with the goal of translating these reagents to clinical trials.  This project will focus on:   1) The development of CRISPR editing reagents with high activity and specificity for ex vivo genome editing in human CD34+ HSPCs.   2) The utilization of unbiased genome-wide techniques for assessing off-target events and other genomic alterations potentially associated with genome editing.   3) The translation of the developed genome editing reagents to clinical application.   These studies will expose the successful candidate to cutting-edge CRISPR/Cas nuclease, base editor and prime editor engineering approaches, and the ex vivo delivery of these reagents to CD34+ HSPCs.  The successful candidate will also learn cutting edge deep-sequencing approaches to evaluate nuclease-based collateral damage to the genome. The successful candidate will have the opportunity to participate in meetings/conferences associated with gene therapy, which will broaden their exposure to cutting edge research within the field. In addition, the successful candidate will learn about Cys2His2 zinc finger protein gene regulation technologies that are being developed in the Wolfe laboratory. Candidates should have a Ph.D. in biology, biochemistry, chemistry or a related biomedical science. In addition, the potential candidate must have strong oral and written communication skills and be eager to present research results at national meetings. GENERAL SUMMARY OF POSITION:   Working as a member of the Medicare Eligibility Enhancement Programs Team at Health Care Finance Solutions (HCFS), the Medicare Premium Assistance Senior Manager is responsible and accountable for oversight of all day-to-day operations of the Medicare Premium Assistance Unit.  The Medicare Premium Assistance Unit is responsible for administering the Massachusetts Medicare Buy-In Program on behalf of the MassHealth program and the Massachusetts Executive Office of Health and Human Services.   Under the direction of the Director or designee, the primary responsibilities of the Senior Manager include the implementation and management of Medicare Buy-In Program policies and procedures, program reporting, enhancement of current program initiatives and supervision of program staff. POSITION SUMMARY:   The GXWT research group focuses on developing and translating viral gene therapy systems to treat rare diseases and other human disorders that currently lack viable treatment options. Our labs collectively focus on developing recombinant adeno-associated virus (rAAV) capsids and vector genome designs to lay the foundation for treating different diseases with or without primary genetic cause. We are also developing rAAV strategies that utilize CRISPR-Cas gene ending, immunotherapies, animal modeling.   Under the general direction of the laboratory PIs, the Veterinary Technician performs various duties related to laboratory animals' care.
